香菇菌丝体杆形病毒的RT-PCR检测及鉴定Detection of a Rod-shaped Virus in Lentinula edodes Mycelium Using RT-PCR

【摘要】 根据GenBank公布的香菇杆形病毒部分基因组序列(登录号:GQ372842)设计引物,建立了香菇菌丝体病毒的反转录-聚合酶链式反应(reverse transcription polymerase chain reaction,RT-PCR)检测方法,浙江省的36个香菇栽培菌株的菌丝体经鉴定,菌株988、939、937、9319、868、66和WL-3的菌丝体不含杆形病毒;菌株灵的菌丝体可能含有杆形病毒;另外28个香菇菌株的菌丝体可能感染杆形病毒。

【Abstract】 Primers designed according to the partial genome sequence(GenBank Accession No: GQ372842) of a rod-shaped virus present in Lentinula edodes mycelium were used to develop a RT-PCR-based viral detection method.Tests conducted on the mycelium of 36 L.edodes cultivars from Zhejiang Province revealed that strains 988,939,937,9319,868,66 and WL-3 were virus-free.Mycelium of 28 other test strains contained the rod-shaped virus but it was unclear if the virus was present in mycelium of L.edodes Ling str...
