【摘要】 对吉林省延吉市和云南省昆明市松茸子实体的ITS序列进行遗传多样性研究。提取两个地区松茸子实体的基因组DNA,应用特异引物对其ITS序列进行PCR扩增及测序,并对云南省昆明市松茸样品进行克隆及测序。之后从GenBank上获取5个地区7条松茸ITS序列,并利用MEGA、BLAST、DNAMAN和ClustalX软件进行对比分析和构建系统发育树。对云南松茸样品克隆发现,ITS序列中有A与G、T与C的颠换,并插入1个碱基T。序列对比发现,松茸ITS序列长度在600 bp~604 bp范围内,G+C含量在43.0%~44.2%之间,共有41个变异位点,ITS2变异位点数要多于ITS1的变异位点数。同时,多个位点存在着转换、颠换、插入和缺失,表明不同产地松茸ITS序列在进化过程中存在差异,为真菌分类、鉴定等研究提供了重要的资料和依据。
